Optimization trays expand a single condition over the X andf Y-axes of a plate, varying 2 of the components. Performing optimizations on previously successful conditions can lead to the discovery of "optimal conditions" for the crystallization of your sample, and larger crystals. The facility sets up the optimization for you, and creates the plate You set up your own optimization, and use the facility to create the plate If considering this route, please be aware of the following:
Optimization Trays
The Crystallization Facility is able to design customized screens based on hits from initial screening with commercial kits. We utilize OptiCrys to design the optimization plate and generate an excel file of liquid values, which is imported into our Tecan Genesis pipetting robot.Options for creating your optimization
This process involves some discussion of the desired optimization parameters, and all pipetting values are setup using OptiCrys in the facility. We then use the facility's robot to pipett the liquids for you, and then setup the crystallization experiment.
Alternatively, you may setup the optimization parameters, create an excel file with all pipetting values, and simply use the facility's robot to pipett the liquids for you. We then setup the crystallization experiment using the optimization plate.
- volumes of each reagent must be greater than 5 ul in all wells
- the robot is unable to pipett high % PEG stock solutions
- OptiCrys does not correctly set up pH gradients
